2. Relationship between immune activation, release of cytokines and formation of neopterin 2.1. Immunological and biological background of neopterin formation

چکیده

Determinations of neopterin levels reflect the stage of activation of the cellular immune system which is of importance in the pathogenesis and progression of various diseases, e.g., in viral infections, in autoimmune or inflammatory diseases, rejection episodes following allograft transplantation and in several malignant diseases. Besides the importance of neopterin as a diagnostic marker new aspects point to a possible biological role of neopterin derivatives (hard copies of this monograph are available at www.brahms.de).

Cellular immune activation, neopterin production, tryptophan degradation and the development of immunodeficiency.

Cellular (Th1-type) immune response is centrally involved in the pathogenesis of various diseases. Within the immunological cascades of Th1-type immunity, interferon gamma (IFN-gamma), among other cytokines, is critically involved. It triggers a series of immune-relevant reactions mostly directed towards forward regulation of the antigen specific immune response. However, in chronic states of i...

Inverse relationship between neopterin and immunoglobulin E.

Polarized human T helper (Th) cells play a key role in the network of the specific immune system compartments. Cell-mediated immune response depends on activation of Th1-type cells, typically producing and releasing interferon-gamma and interleukin-2, whereas activation of Th2-type cells and production of cytokines such as interleukin-4, -5, and -10 are involved in humoral immune response and t...

Analysis of urinary pterins in autism.

The pterins, neopterin and biopterin, occur normally in body lluids including urine. It has been known for some time that increased production and release of neopterin and 7,sdihydroneopterin accompanies immune activation of macrophages both in vilro and in vivo [ 1.21. Interferon gamma and tuniour necrosis factor alpha are the key cytokines which lead to this increase in neopterin levels [2] b...

A non-specific biomarker of disease activity in HIV/AIDS patients from resource-limited environments.

BACKGROUND A general non-specific marker of disease activity that could alert the clinician and prompt further investigation would be of value in patients with HIV/AIDS, especially in resource limited environments. OBJECTIVE To investigate the potential of neopterin as non-specific biomarker in patients with advanced HIV/AIDS. METHODS Cross-sectional study in 105 HIV positive patients (75 o...

ENZYME-LINKED IMMUNOSORBENT ASSAY OF NEOPTERIN USING PENICILLINASE AS LABEL

An enzyme-linked immunosorbent assay for neopterin using penicillinase as marker enzyme is reported here by polyclonal antibodies against neopterin conjugated to bovine serum albumin which were raised in rabbits. Immunoglobulin fractions were purified and coated on wells of microtiter plates. A chain heterology was introduced in neopterin derivative and conjugated to penicillinase. The assa...
